Short Description:
Polyphenol oxidases (PPOs) are a group of copper-containing enzymes that catalyze the o-hydroxylation of monophenols to o-diphenols, as well as the oxidation of o-diphenols to quinones in the presence of oxygen. To better understand the functional role of the single PPO enzyme in walnut, we used RNA interference (RNAi) to generate a series of transgenic walnut lines with greatly reduced PPO activity (PPO-silenced). Multiple leaf tissue samples were collected from wild-type trees and from PPO-silenced trees. Three independent wild-type biological replicates and three independent PPO-silenced biological replicates were made using a TruSeq RNA kit. Illumina HiSeq2000 was used for sequencing of RNA isolated from the samples.
